"Our team is divided into 3 subgroups, each followed up on an analytical chemistry of another focus. The first of the subgroups solves the preparation and characterization of hyaluronan oligosaccharides and degradation products of hyaluronan derivatives. In this work, we solve analytical problems related to preparative low- or high-pressure chromatography, separation and identification of positional isomers present in the complex enzymatic hydrolyzate of the derivative, etc.
The second subgroup in the company develops analytical chemistry as such. It introduces, verifies and validates analytical methods suitable for the identification and quantification of impurities in prepared products and, last but not least, performs selected routine determinations. In this work, the subgroup works closely with the control analytical laboratory and provides analytical support to all R&D colleagues and actively participates in solving problems related to the production of cosmetics, pharmaceuticals and final products of the company.
Colleagues from the last subgroup are actively involved in conducting and evaluating the pharmacokinetics of our products (ADME study - adsorption, distribution, metabolism, and excretion), preceded by the careful development of analytical methods focusing on the quantification of substances in biological matrices. In addition, we also focus on the molecular imaging of substances in cuts of selected tissues. Our entire group cooperates with a wide range of basic research departments.”
Mgr. Martina Hermannová Ph.D., Head of Pharmacokinetics Team
Methods and technical background:
For the separation and quantification of impurities and degradation products, we use modern HPLC, UHPLC, and GC devices, always in combination with a diode array detector, an evaporative light scattering, refractometric or conductivity detector and/or mass spectrometer. To determine the molecular weight of hyaluronan, we use combinations of gel permeation chromatography with a light scattering detector (so-called GPC-MALLS). For chemical characterization of our derivatives and positional isomers of modified oligosaccharides of hyaluronan, we mainly use NMR and LC-MS. Quantification of analytes in biological matrices for pharmacokinetic studies is performed on LC-MS based on triple quadrupole and molecular imaging by desorption electrospray.
